The author first presents the use of the [1+2+3] one-pot sequential strategy to successfully synthesise the RM2 antigen and its analogues as single stereoisomers in every glycosylation step, producing good yields and stereoselectivity. He then introduces the conjugation of the synthetic RM2 antigen to the carrier protein CRM197 in an average number of 1?10 to create the prostate cancer vaccine candidate, which is combined with ?-galactosylceramide C1, its analogue C34, or Alu. The results of the vaccination studies in mice are also described and indicate that the strongest anti-RM2 antigen titer is exhibited when one molecule of diphtheria toxin (DT) is conjugated with an average of 4.7 molecules of RM2 antigen (DT-RM4.7) and adjuvanted with the glycolipid C34. More importantly, the induced mouse antibodies mediate the effective complement-dependent cytotoxicity (CDC) against the prostate cancer cell line LNCap. The study presented in this thesis is the first ever to successfully synthesize this complex glycan molecule. Owing to the steric hindrance of the adjacent sialyl moiety, the introduction of two sialic acid units to the compact and rigid 3,4 di branched galactoside unit is very challenging and the ?-selective and efficient glycosylation of the galactosamine moiety at the 4-position of di branched galactose is also problematic.
["dodax
undefined
synthesis-and-vaccine-evaluation-of-the-tumor-associated-carbohydrate-antigen-rm2-from-prostate-cancer
/unterhaltung/produkt/synthesis-and-vaccine-evaluation-of-the-tumor-associated-carbohydrate-antigen-rm2-from-prostate-cancer/
["orell-fuessli

A number of standards and techniques have been developed by the World Wide Web Consortium (W3C), e.g., the Resource Description Framework (RDF), which provides a general method for conceptual descriptions for Web resources, and SPARQL, an RDF querying language. Recent examples of large RDF data with billions of facts include the UniProt comprehensive catalog of protein sequence, function and annotation data, the RDF data extracted from Wikipedia, and Princeton University?s WordNet. Clearly, querying performance has become a key issue for Semantic Web applications. In his book, Groppe details various aspects of high-performance Semantic Web data management and query processing. His presentation fills the gap between Semantic Web and database books, which either fail to take into account the performance issues of large-scale data management or fail to exploit the special properties of Semantic Web data models and queries. After a general introduction to the relevant Semantic Web standards, he presents specialized indexing and sorting algorithms, adapted approaches for logical and physical query optimization, optimization possibilities when using the parallel database technologies of today?s multicore processors, and visual and embedded query languages.Groppe primarily targets researchers, students, and developers of large-scale Semantic Web applications. On the complementary book webpage readers will find additional material, such as an online demonstration of a query engine, and exercises, and their solutions, that challenge their comprehension of the topics presented.
["dodax

Of course, existence of preferential orientation of the absorbing dipoles in the tubuli of the rhabdomeres can not be totally rejected. We hope that the concept of the "universal" photoreceptor membrane may serve as the useful instrument when dealing with newly discovered properties of visual cells so that true mechanisms of electrical and optical coupling will be searched for instead of assumptions being made on additional properties of the photoreceptor membrane in every new animal under study. 5. Absorption Spectrum of the Universal Photoreceptor Membrane and Spectral Sensitivity of the Photoreceptor 5. 1 Preliminary Notes It seems nearly self-evident that the absorption spectrum of the pho toreceptor membrane coincides exactly with that of the visual pigment it contains. Hence, the membrane must exhibit three bands of absorp tion - the principal band with its peak within the limits of visible spectrum (or a-peak), the secondary band between 340 and 380 nm (S peak), and the third, protein band, in the ultraviolet (UV) at 280 nm (COLLINS et al. , 1952). The main peak of absorption is located within the range 433-575 nm for retinol-based pigments and between 438 and 620 nm for 3-dehydroretinol-based pigments, the position of Amax de pending on many ecological factors.
["dodax

Based on this definition, the field of computational systems biology has been in existence for some time. However, the recent confluence of high-throughput methodology for biological data gathering,genome-scalesequencing,andcomputationalprocessingpowerhasdrivena reinvention and expansion of this field. The expansions include not only modeling of small metabolic (1–3) and signaling systems (2, 4) but also modeling of the relati- ships between biological components in very large systems, including whole cells and organisms (5–15). Generally, these models provide a general overview of one or more aspects of these systems and leave the determination of details to experimentalists focused on smaller subsystems. The promise of such approaches is that they will elucidate patterns, relationships, and general features, which are not evident from examining specific components or subsystems. There are many successful applications of CI in many sub elds of biology, including bioinformatics, computational - nomics, protein structure prediction, or neuronal systems modeling and an- ysis. However, there still are many open problems in biology that are in d- perate need of advanced and e cient computational methodologies to deal with tremendous amounts of data that those problems are plagued by. - fortunately, biology researchers are very often unaware of the abundance of computational techniques that they could put to use to help them analyze and understand the data underlying their research inquiries. On the other hand, computational intelligence practitioners are often unfamiliar with the part- ular problems that their new, state-of-the-art algorithms could be successfully applied for. The separation between the two worlds is partially caused by the use of di erent languages in these two spheres of science, but also by the relatively small number of publications devoted solely to the purpose of fac- itating the exchange of new computational algorithms and methodologies on one hand, and the needs of the biology realm on the other. The purpose of this book is to provide a medium for such an exchange of expertise and concerns. In order to achieve the goal, we have solicited cont- butions from both computational intelligence as well as biology researchers.
["weltbild","orell-fuessli
